Urine Pregnancy Test can be done even on day one of the missed periods.. however it is not necessary that it is positive... so if you are expecting pregnancy repeat the test after one week... if you are overdue.. or else you can do a beta HCG blood test which is a confirmatory test

Urine pregnancy test is positive around the time of missed period. It is advisable to repeat test after one week of missed period if you still do not get period before taking any further course of action
