You should do semen analysis after two days of abstinence from sex. If your semen report comes normal then your wife's tests will need to be done, at first Hb TC DC TSH SUGAR PP PROLACTIN and Ultrasound of Pelvis. Depending on these reports treatment will be given. If your semen report is not normal then first you will be given treatment for 3 months.

Do NOT lose hope. Ask her to start folic acid. Try regularly at least 2-3 times a week. As 2 years have passed, you should contact doctor and start testing (semen test for husband, and for wife- HSG and blood tests for AMH, Rubella IgG and Thalassaemia).
